报告人简介:

Dr. Su'etsugu has received his PhD from Kyushu University, Japan in 2005, and continued his work on chromosome replication of Escherichia coli, as Assistant Professor at the same university. He then joined Jeff Errington's lab as a postdoctoral researcher at Newcastle University, UK. After moving back to Japan, he became an Associate Professor at Rikkyo University, Tokyo in 2013, and started his laboratory. The laboratory currently focuses on synthetic biology based on in vitro reconstitution of bacterial self-replication systems. He is also the co-founder and Chief Scientific Officer (CSO) of a spin-off company of his DNA assembly and amplification technology, OriCiro Genomics, Inc.


报告内容:

Propagation of genetic information is a fundamental biological property that is common to living organisms. Here, we have reconstituted an entire replication cycle of the Escherichia coli circular chromosome with 25 purified proteins that catalyze initiation, fork progression, Okazaki-fragment maturation, and decatenation (Su'etsugu et al., 2017 NAR). Because the decatenation provides supercoiled monomer DNA that are competent for the next round of replication initiation, the replication cycle repeats autonomously and continuously. This Replication-Cycle Reaction (RCR) thus propagates large circular DNA molecules exponentially in an isothermal condition. The E. coli transcription and translation reaction has been also reconstituted in vitro as PURE system (Shimizu et al., 2001 Nat. biotech.). I will talk about our approach towards self-replication reaction by integration of RCR and PURE system.
