报告人简介:Boyi Gan, Ph.D,美国德克萨斯大学MD安德森癌症中心,放射肿瘤学系,Assistant Professor。2000年获得复旦大学硕士学位,2006年获得美国康奈尔大学比较生物医学博士学位,2006-2010年在美国哈佛大学从事博士后研究,2010-2011年,哈佛医学院讲师。2003年至今,美国细胞生物学会会员,2006年至今,美国癌症研究协会会员;Boyi Gan教授还在Nature, Cancer Cell, Journal of Biological Chemistry ,Genes and Development等国际著名期刊上发表学术论文几十余篇。
报告内容简介:Hematopoietic stem cells (HSCs), the best understood adult stem cells, function to sustain all blood lineages. Using HSC as a stem cell model system, we have sought to understand the role of tumor suppressors involved in cancer metabolism in the regulation of stem cell homeostasis, including TSC, a key negative regulator of mTORC1 activation and cell growth, and LKB1, a critical component controlling energy metabolism. Specifically, we demonstrate that somatic deletion of tumor suppressors TSC1 or Lkb1 in adult mouse produces striking stem cell and derivative effector cell phenotypes characterized by increased HSC cell cycling, marked HSC depletion, defective long-term repopulating potential, and developmental aberrations in various hematopoietic lineages. Together, these studies have delineated an intimate link between tumor suppressor pathways that control cancer metabolism and those that regulate stem cell homeostasis, and pointed to the critical importance of coupling energy availability and tissue homeostatic demands.
